光駆動イオンポンプにおける構造と機能相関

研究概要

本研究では光駆動プロトンポンプであるバクテリオロドプシンと光駆動塩素イオンポンプであるハロロドプシンにおいて光反応に伴う蛋白質の構造変化が機能とどのように相関するのか明らかにすることを目的とした。特に偏光法、時間分解法といった新たな手法を赤外分光法に導入し、水分子の角度情報や低温では捕捉できないような中間体の構造解析を目指した。その結果本年度は以下の成果が得られた。
(1)バクテリオロドプシンの構造解析:膜蛋白質におけるプロトンポンプには水分子が関与することが考えられるが、我々はこれまでに赤外分光法を用いて蛋白質に結合した水分子の水素結合変化を直接、観測することに成功している。このような水分子が蛋白質内部の制限された空間にどのように存在するのか明らかにするために偏光FTIR分光法を用いた実験を行ったところ、3643cm^<-1>に伸縮振動をもつ水分子はその双極子モーメントが膜の法線に対して60℃の角度を成すことがわかった。水分子は蛋白質内部で特定の配向をしており、その変化がプロトンポンプをもたらすことが示唆された。
一方、新たに導入した時間分解赤外分光法から低温では捕捉できないO中間体を捉え、特有の蛋白質部分の構造変化を明らかにした。さらに発色団レチナ-ルの変化から遅延した蛋白質部分の変化を見出した。4ナノメーター程度離れた膜の間をプロトンを輸送するためには協奏的な蛋白質の構造変化が要求されるが、このような時間的な遅延は構造変化の因果関係を考察する上で重要な知見を与えるものと考えられる。
(2)ハロロドプシンの構造解析:低温赤外分光法によりハロロドプシンの構造解析を行い、L中間体に2種類の構造体が存在することを見出した。
