研究概要

地球上には92種類を超える元素が存在する。生命体はこれらの元素の中から約30種類の元素を選択的に利用している。長い進化の過程において生命体による金属イオンの選択は,そのときどきの環境に応じて変化してきた。今から約90年前に脊椎動物ホヤの血球から高濃度のバナジウムが検出された。その生理学的意義はいまだ不明な点が多いが,近年系統学的に遠い位置にある環形動物エラコに高濃度のバナジウムが濃縮されていることがわかり,生物界にあまり例を見ないバナジウムの選択的利用が脚光を浴びることとなった。
本研究では,脊椎動物と環形動物に共通のバナジウム濃縮機構があるか否かを解析することを目的として,次のような成果を得た。
(1) バナジウム結合タンパク質(VAP)からバナジウムを除去したアポタンパク質を作製し再結合実験をしたところ,10原子のバナジウムが結合することを見いだした(未発表)。
(2) バナジウム結合タンパク質(VAP)のうち,12.5kDaと15kDaをコードする遺伝子の配列を明らかにした(未発表)。
(3) VAP遺伝子を発現ベクターに組み込み融合タンパク質を合成した(未発表)。
(4) VAP遺伝子をプローブとしてサザンプロット解析を行うために,エラコのゲノムDNAの抽出を試みた。
