古細菌サーモプラズマの細胞骨格と脂質合成系の研究

研究概要	16S-rRNAの塩基配列及びいくつかの蛋白質の塩基配列に基ずく系統樹から真核生物のは古細菌から派生してきたことが明らかとなって来つつある。その過程で、好気性細菌が細胞内共生してミトコンドリアとなり、シアノバクテリアが細胞内共生して葉緑体となったと考えられている。古細菌から真核生物が形成される過程に関する情報を得る目的で、好熱好酸性の古細菌Thermoplasmaを材料として研究を行った。古細菌は通常蛋白質の2次元結晶状の構造を持つS-層あるいは真正細菌のペプチドグリカンに類似の構造シュウドムレインの細胞壁を持っている。それに対して、Thermoplasmaは細胞膜の外にはまったく構造を持たない。現在、Thermoplasmaが直接の祖先であるかどうかについては必ずしもはっきりしていないが、Thermoplasmaが細胞壁を持っていないことから、細胞内骨格を持っている可能性は大きい。この菌はpHをアルカリにすることで溶菌することが知られている。そこで、アルカリで処理した後に遠心分画を行い、沈澱の構成蛋白質をSDS-PAGE及び原子間力顕微鏡を用いて分析した。アルカリ処理によって得た沈澱は膜標品であると推定された。一方細胞を非イオン性の界面活性剤Triton-X100で処理したところ、細胞膜が除去された、凝集体が観察された。一方、Thermoplasmaの膜脂質はテトラエーテルとジエーテルによって構成されているが、その極性基に関する分析は行われていない。そこでThermoplasmaの極性脂質をTLCで分析する条件を検討した。Thermoplasmaの脂質合成を合成阻害剤テルビナフィンで阻害したところ、いくつかの前駆体と予想される脂質が検出された。
