植物病原糸状菌におけるトランスポゾンタギング

研究概要	トランスポゾンは、各種生物における遺伝子単離の強力なツールとして利用されている。いわゆるトランスポゾンタギングである。申請者らは、ユウガオつる割病菌(Fusarium oxysporum f.sp.lagenariae)から、これまで病原糸状菌では全く報告されていなかったAcタイプトランスポゾン様配列(Tfolと命名)を単離した。本研究では、Tfol配列を用いた糸状菌の遺伝子タギング法の確立を目指し、以下の研究を行った。 1.Fusarium oxysporumの各種分化型におけるTfo1様配列の分布 F.oxysporumの各種分化型におけるTfo1様配列の分布をサザンハイブリダイゼーションとPCR法を用いて調査した。その結果、ユウガオつる割病菌だけでなく、用いた10分化型のすべての菌株からTfo1様配列が検出され、本配列がF.oxysporumに広く分布していることが明らかとなった。 2.Tfo1からの転写RNAの検出 Tfo1にはトランスポゼ-ス様タンパク質の読み枠(ORF1)が存在する。ユウガオつる割病菌とトマト根腐萎ちょう病菌(f.sp.radicis-lycopersici)について、ノーザンハイブリダイゼーションとRT-PCR法を用いてORF1の転写RNAの検出を試みた。その結果、どちらの分化型からも転写RNAが検出され、ORF1がトランスポゼ-ス遺伝子として機能していることが示された。以上の結果は、Tfo1がトランスポゾン活性を有すること、さらに糸状菌のトランスポゾンタギングに利用できることを強く示唆した。そこで現在、Tfo1配列を組み込んだ形質転換ベクターを各種糸状菌に導入し、遺伝子タギングを試みている。
