胚性幹細胞の生殖系列細胞へのin vitro分化誘導に関する基礎研究

研究概要	胚性幹細胞(ES細胞)や胚性生殖細胞(EG細胞)は、遺伝子ターゲティング以外にもさまざまな応用が応用動物科学分野で試みられている。現在のところ、ES細胞から生殖系列細胞を確立した報告は無いが、適当な培養条件でES細胞をin vitroで生殖細胞系列化することができれば、その応用動物科学上の意味は極めて大きい。本研究の目的は、マウスES細胞をin vitroでPGCに分化誘導する条件を確立し、さらに、得られた結果をもとに、家畜のES細胞、ないしはES-like細胞について同様の実験を試み、将来の実用化への基礎を確立することにある。この目的に向けて、本年度は、単一のES細胞を胚盤胞に顕微注入して、その分化能、特に、生殖系列への分化能を解析した。その結果、単一のES細胞が生殖系列、および、解析を行った限りにおける全ての体細胞系譜に分化可能であることを確認した。さらに、ES細胞のfeeder細胞として、従来から用いられているembryonic fibroblastsの他、yolk sac細胞をfeeder細胞として用い、ES細胞(R1株)の培養を試みた。PGCのマーカーとして、しばしば用いられる4C9抗体の他、2C9など複数のPGC特異的抗体を用いて、さまざまな条件下で培養したES細胞を蛍光抗体法により解析した。特にXXの核型をもつES細胞について検討を行った。ES細胞の中でも、移動性の細胞に4C9陽性細胞が多いことが確認された。さらに、マトリゲル上で培養したES細胞のコロニーの中に、コロニー全体が4C9、2C9に対して陽性反応を示すものがあることが確認され、ES細胞の生殖系列細胞化に、興味深い予備的結果が得られた。これらの知見を端緒として、現在さらに研究を進めている。
