大腸癌においてサイトカインにより誘導されるFas抗原とアポトーシスの解析

研究概要	IFNとTNFまたは抗癌剤である5-FUの併用時の、大腸癌細胞株COLO201におけるFasの発現とアポトーシス誘導を検討した。また、アポトーシスの起こるメカニズムとしてBcl-2やその関連タンパクの関与に関しても検討を加えた。 1. IFN-γとTNF-αは単独でも濃度依存性または時間依存性に大腸癌細胞株であるCOLO201のFasの発現を増強し、またアポトーシスを誘導するが、両者の併用が、それぞれの単独投与より抗腫瘍効果があり、さらによりFas抗原の発現を増強する。また、このFas抗原を刺激することで腫瘍細胞が容易にアポトーシスに陥ることが明らかとなった。アポトーシスに陥った腫瘍細胞ではBcl-2の発現低下Baxの発現増強がタンパクレベルまたmRNAレベルでみられ、腫瘍細胞がアボトーシスに陥るメカニズムとしてはBcl-2の発現低下とBaxの発現増強が関与していると考えられた。 2. IFN-γと大腸癌に用いられる抗癌剤である5-FUとの併用に関しても検討を加えた。5-FU単独でアポトーシスを誘導可能であるが、IFN-γを加えることで相乗的にアポトーシス細胞が増加し、このとき、Bcl-Xの発現に変化はみられなかったが、Bcl-2の発現低下、Baxの発現増強が認められた。 3. 5-FUとIFN-γ投与時のCaspaseの活性化ても検討を加えた。5.FUとIFN-γは単独でcaspase3と8の活性化を誘導したが、両者の併用でさらに活性化が認められた。これは、基質を用いた発色法とwestern blotingにて確認された。
