新規のケモカイン・ファミリー分子の分子生物学的方法による検索

研究概要

すでにクローニングしているヒト・ケモカイン・レセプター様遺伝子HM74のリガンドの精製を目的として、HM74の細胞外領域とglutathione-S-transferase(GST)との融合蛋白を発現し、この蛋白を固相化したアフィニティ・カラムを本年度作成した。現在、このカラムを用いて、リガンドの精製を試みている。また、HM74の種々の細胞での発現パターンを検討するために、作成した融合蛋白を免疫源として抗体を作成している。マウスPBSFをブロープとした、cosmidライブラリーのスクリーニングでは陽性クローンを現在のところ得ることができないために、他の方法によるスクリーニジグも検討している。
以上の研究と並行して、インターロイキン8(IL-8)・macrophage inflammtory protein(MIP)-1α・RANTESに関する研究を進め、以下の結果を得た。
1)エンドトキシン誘発による急性呼吸窮迫症候群・再灌流による脳傷害が、抗IL-8抗体によって軽減されたことから、これらの好中球依存性におきる組織傷害の発生にIL-8が密接に関与していることが示唆された。
2)フォスファターゼ阻害剤が、Erk-1の活性化を引き起こすとともに、IκBαのリン酸化と蛋白分解、ひいてはNF-κBの活性化を引き起こすことによって、IL-8遺伝子転写を引き起こすことを明らかにした。
3)骨髄赤芽球系細胞表面にはMIP-1αに対するレセプターであるCCR1が発現しており、MIP-1αはCCRlと結合して、赤芽球系細胞の分化を抑制することを明らかにした。
4)好酸球性肺炎の肺胞洗浄液中で、RANTESが特異的に増加していて、これが肺への好酸球浸潤に関与している可能性を示唆する結果を得た。
