川崎病の病因の遺伝子学的な独創的手法による解明とその展開

研究概要	本研究の目的はユニークな手法を用いて川崎病の病因を明らかにすることである。平成10年度は (1) 川崎病患者でみられる抗体をコードしている免疫グロブリン遺伝子を決定するために川崎病患者末消リンパ球からEpstein-Barrウイルスを用いてB細胞株を樹立した。さらにlimited dilution法によりクローニングを試みた。B細胞株や末梢リンパ球から独立してDNAおよびRNAを採集し、RNAからはcDNAを作成した。DNAまたはcDNAを用いて、免疫グロブリン遺伝子のvariable regionのVDJとconstant region のCγ部分のprimerを設定して、PCRまたはRT-PCRを用いて増巾させ、増巾バンドをPC19ベクターにてクローニングした。これから川崎病において使用されているV領域,CDR3領域の塩基配列の特性を決定し、さらにアミノ酸配列を決定した。 (2) 川崎病におけるV領域のアミノ酸配列からコンピューターを用いて系統樹を作成したところ予想に反して幅広く分布していたが、一部に集積の傾向が認められた。この集積性が他の既知の抗原に対する抗体タンパクのV領域といかなる差異を有するか更に検討をすすめており、興味ある結果が得られつつある。以上の結果をもとに現在、(1)決定された塩基配列を有するVDJ-Cγを発現ベクターに組み込んで大腸菌でタンパク、すなわち抗体を作成し(2)さらに作成した川崎病特異抗体を用いた検索から川崎病の病因を確定するためカラムによる抗体吸着と(3)既存の抗原に対比してV領域アミノ酸をコンピューター分析しており、これらから結論を得る。
