研究概要

本年度は、マウスにおけるproinsulinのbioavailabilityの検討とproinsulin遺伝子を組み込んだRetrovirusを産生する、producer細胞の作製を行った。
まず、マウスにおいて血糖を低下させるのに必要なプロインスリン量を求めるため、骨髄移植のrecipientとして想定しているC57Bl/6mouseにipITTを施行した。Human proinsulin 350nmol/kg投与により、Human insulin 7.0nmol/kgと同程度の血糖降下作用を認めたことから、human proinsulinはhuman insulinに対し、1/50程度のbioavailabilityを保持することが明らかになった。
次に、proinsulin遺伝子を組み込んだRetrovirusを産生する、producer細胞の作製に着手した。ヒト膵組織よりRT-PCR法によりpreproinsulin全翻訳領域のmRNAを増幅し、cDNAを作成した。また、コントロールとして、このcDNAよりA-chainをdeleteした構造のcDNAを作成した。次に、moloney白血病ウイルス由来のレトロウイルスベクターpGK,pRxのLTR下流域にHINおよびHINcのconstructを挿入した合計4種類のretrovirus vectorを作成した。このproducer細胞のgenomeを抽出し、PCR法により、挿入したconstructが組み込まれていることを確認した。
