悪性グリオーマ普遍発現遺伝子の検索とそのプロモーターを用いた組織特異的遺伝子治療

研究概要

悪性グリオーマに対する特異的遺伝子療法を開発する第一段階として、、ミエリン塩基性蛋白(MBP)遺伝子発現プロモータで殺細胞遺伝子を制御したレトロウイルスベクターを作製した(J Neurosci Res 36:472-479,1993)。しかし、そのプロモータ活性は、モロニ-白血病ウイルスのLTRやSV40プロモータなど組織非特異的プロモータと比較すると弱い。それ故、新しい遺伝子スクリーニング法であるSAGE(Serial analysis of gene expression)法を用いて、悪性グリオーマ普遍的発現遺伝子およびそのプロモータを検索し、より効率のよい遺伝子治療法の開発を計画した。今年度は、悪性グリオーマ患者の手術摘出組織やヒト悪性グリオーマ細胞株よりメッセンジャーRNA(mRNA)を抽出し、リバーストランスクリプターゼ(RT)法を用いてcDNAを合成した。さらに、poly A付加部位より9塩基のTagを作製し、これらを縦一列に配列した合計約500塩基からなるDNAを合成した後シークエンスを行い、グリオーマ手術摘出組織や各種細胞株より最も普遍的に発現している遺伝子をヒトゲノムデータベースより検索している段階である。今後は、悪性グリオーマ普遍的発現遺伝子を幾種類か決定し、それが既知のものであればその遺伝子プロモータを用いて、また未知の遺伝子であればそれをクローニングし、その遺伝子のプロモータ領域を用いて、1acZ遺伝子を制御する組換えベクターを作製する予定である。そして、このベクターを用いて、各ヒトグリオーマ細胞株におけるプロモータ活性を検討し、その中で一番活性の高いプロモータを用いて殺細胞遺伝子(HTK)を制御できるベクターを構築し、悪性グリオーマ特異的遺伝子治療の新たな基礎的研究を行う予定である。
