研究概要

1. ヒト膀胱排尿筋における脱感作機構の研究-G蛋白質共役受容体リン酸化酵素の発現と薬理学的検討
方法
1) 免疫組織学的解祈:ヒト膀胱を用いて、GRK2に対する抗体による免疫組織学的検討を行った。
2) 分子生物学的解析:ヒト膀胱から全RNAを抽出し、ムスカリンM2、M3受容体、GRK2に対するreverse transcription/polymerase chain reaction(RT/fPCR)を行った。
3) 薬理学的検討:KClによる収縮を前収縮とし、カルバコール投与後のKCl収縮の変化から脱感作の検討を行った。
結果
ヒト膀胱ではGRK2のmRNA及び蛋白の発現を認めた。 機能的にも、カルバコール投与後のKCl収縮の変化から脱感作を認めた。
2. ヒト正常、閉塞、神経因性膀胱排尿筋におけるG蛋白質共役受容体リン酸化酵素の発現
方法
1) 免疫組織学的解祈:ヒト正常、閉塞、神経因性膀胱排尿筋を用いて、GRK2、GRK3、GRK4に対する抗体による免疫組織学的検討を行った。
結果
正常群では。GRK2は排尿筋のみに陽性、GRK4は血管のみに陽性であり、GRK3は染色されなかった。
閉塞群では、GRK2は排尿筋に陽性、血管にわずかに染色を認め、GRK3、GRK4は染色されなかった。
神経因性膀胱群では、GRK2は排尿筋に陽性、血管にわずかに染色を認め、GRK3は排尿筋のみ、GRK4は染色されなかった。
