研究概要

常染色体劣性無精子症の原因候補遺伝子であるヒトのDAZLA(Deleted in Azoospermia-like autosomal)遺伝子を解析することにより以下の成果を得た.
1. ヒトDAZLA遺伝子の構造・染色体局在の決定
1)ヒトDAZLA遺伝子はY染色体でコードされるヒトDAZ遺伝子と同一のエクソン・イントロン構造を有していること,2)DAZLA遺伝子は第3染色体のバンドp25領域に存在すること,が明らかになった.
2. ヒトDAZLAタンパク質に特異的な抗体の作製
RNA結合タンパク質と考えられるDAZLA分子は,核,サイトゾル,あるいはその両方に存在する可能性がある.大腸菌で発現された組み換えヒトDAZLA分子でラビットを免疫することにより,DAZLAタンパク質に特異的な抗体を作製した.本抗体を用いてDAZLAタンパク質の細胞内局在を検討したところ,DAZLA分子は主としてサイトゾルに存在することがあきらかになった.DAZLA分子は主に精原細胞・精母細胞において発現されている.
3. DAZ/DAZLA遺伝子の分岐年代の決定
ヒトのゲノムにはY染色体でコードされるDAZ遺伝子と常染色体でコードされるDAZLA遺伝子の両方が存在するが,マウスのゲノムには常染色体でコードされるDazla遺伝子しか存在しない.本研究により,DAZは旧世界ザルと新世界ザルが分岐した後に前者の系統においてY染色体に転座された遺伝子であることが明らかになった.
4. 本遺伝子が常染色体劣性無精子症の原因遺伝子であるか否かは,現在なお不明である.
