切断部位特異抗体を利用したアポトーシスプロテアーゼの標的分子の探索

研究概要	研究代表者が既に確立した切断部位特異抗体(プロテアーゼによって限定分解を受けたポリペプチド鎖末端領域に対する抗体)の手法を利用して、アポトーシスの情報伝達にかかわるプロテアーゼ(カスパーゼ)の未知の標的分子を同定する方法の開発を試みた。現在、カスパーゼは10種類以上に関連遺伝子が発見され、アポトーシスの実行過程において連鎖的に活性化すると考えられている。アポトーシス誘導時には様々なタンパク質がカスパーゼで限定分解を受けると、その生理的意義は明らかではない。いずれのカスパーゼも標的タンパク質のアスパラギン酸残基のカルボキシル末端側でペプチド結合を開裂する。そこでハプテンとして、10残基のププチドCys-X_8-Asp(CX_8D)を化学合成し、ウサギで切断部位特異抗体を作成した。ここでXはCys以外の19種類のアミノ酸の混合物であり、配列をランダム化してできるだけ多くの種類の抗原(アスパラギン酸残基のカルボキシル末端をもつポリペプチド)に結合するような抗体の集団がでるることを期待した。予備実験として既に切断部位特異抗体を得ているカルパインやプロテインキナーゼCについて、固定化した抗体で細胞粗抽出液から抗原を捕捉する方法を検討した。この結果に基づいてCX_8Dに対する抗体に結合するタンパク質をアポトーシスを起こした細胞でアフィニティーカラムを用いて検索すると、分子量3万のピリペプチドが見つかった。これはポリ(ADPリボース)ポリメラーゼに対する抗体と交叉したので、本酵素がカスパーゼ3で切断された断片と考えられる。未知の基質タンパク質を同定すべく、さらに微量の抗原を検索している。
