イモリにおける遺伝子発現とゲノム上の遺伝子の分布

研究概要

本研究ではゲノムが特に大きい両生類であるアカハライモリ(Cynops pyrrhogaster)を対象として、生物におけるゲノム情報構造とその発現の仕組みに関する新しい知見を得ることを目的とし,そのための第一歩としてこの生物のゲノム構造の一端を解析するべく企画立案した。
過去2年間、まず(1)野外からアカハライモリの幼生を多数採集して凍結保存し、それらの幼生の細胞からイモリのcDNAを抽出してクローンバンクを作成し,得られたクローンの塩基配列を決定することにより,イモリで実際に発現している遺伝情報をできるだけ多く解析して他の生物種と比較することを試みた。しかし、イモリからのDNA抽出やcDNA作製の過程で予期しなかったようないろいろな問題が生じて手間取り、あまり多くのcDNAクローンについての解析が進められなかった。またこれと平行して,(2)イモリの染色体DNAのYACクローンバンクを作成し、両者の組織的なハイブリッド形成を調べることにより、発現している遺伝子のYACクローン上の分布を詳細に解析することも計画した。この線に添って一応かなりの数のYACクローンは作製できたが、cDNAクローンの方に問題があって、それらの解析はあまり進んでいない。さらに,(3)できるだけ多くのcDNAについてそのゲノムDNAクローンを作成し,エキソンやイントロンの解析を進める予定であったが、これについても同様の理由であまり進められなかった。
今後、方法を改善してcDNAクローンバンクの作製を進め、本研究で得られているYACクローンを用いて当初予定した解析を継続的に進めていく予定である。
