真核細胞の転写-修復共役反応および複製-修復共役反応の無細胞系における解析

研究概要	研究代表者および研究分担者は、真核細胞における転写と修復、および複製と修復の各共役反応の無細胞系の構築とその解析を目指して、以下に述べる研究を行った。 1)既に我々が確立している、紫外線照射したSV40ミニ染色体を鋳型とする無細胞DNA修復系をベースとして、無細胞転写系に必要な成分を加えたり、あるいは反応条件を転写反応のそれに近付けたりして、転写と修復の共役反応を起こさせるように工夫した。しかしながら条件をどのように振っても、転写と修復の両反応を効率よく起こさせることは出来なかった。 2)1)で述べたように、solubleな条件での転写と修復の共役系の構築は困難であったので、この共役には何らかの細胞構造が必要ではないかと考え、分離核での転写-修復共役反応の確立を試みた。特異的阻害剤としてα-アマニチンを用いることにより、HeLa細胞からの分離核系での転写産物にRNAポリメラーゼII(Pol.II)によるものが含まれていること、そして紫外線照射したHeLa細胞から調製した核では、Pol.IIによる転写産物が増加することを見出した。一方、transcription-coupled repairに異常を持つことが知られているコケイン症候群患者細胞からの分離核では、紫外線処理によってPol.IIによると思われる転写産物が消失した。これらのことは、この分離核系が転写-修復共役反応を見るためのよいモデル系であることを示している。 3)複製と修復の共役反応については、SV40ウイルスのオリジンを有するプラスミドに1カ所損傷を持たせたものを鋳型として、複製と修復を共役させる試みを始めた。
