イオンチャネル型シグマI受容体の内在性リガンドの探索

研究概要	シグマI受容体の内在性リガンドの探索に先立って、マウス脳各部位のシナプス膜標品を用いて高親和性GTPase活性あるいは〔^<35>S〕GTP_γS結合活性を測定したところ、GTPase活性は、シグマ受容体アゴニストの(+)-pentazocine、(+)-3-PPP、SA4503などによって有意な刺激作用がみられ、アンタゴニストのNE-l00はそれらの刺激効果を有意に抑制した。また、〔^<35>S〕GTP_γS結合活性もGTPase活性の場合と同様にアゴニストによる活性上昇作用が認められた。さらに、小脳シナプス膜での(+)-pentazocineによる〔^<35>S〕GTP_γS結合活性上昇作用は百日咳毒素処置により遮断され、リコンビナントのGi蛋白質を再構成することで活性の回復が認められた。以上のことから、G蛋白質連関型のシグマ受容体が存在することを明らかとした。しかしながら、脳各部位における〔^3H〕(+)-pentazocineの受容体結合活性と〔^<35>S〕GTP_γS結合活性との間に相関性はみられず、G蛋白質と連関しないシグマ受容体の存在も示唆された。近年、シグマI受容体がクローニングされ、マウス小脳からRT-PCR法によって得られた受容体クローンをバキュロウイルス/Sf21細胞発現系に組み込み、その受容体と各種リコンビナントG蛋白質を特異的に共発現させた細胞膜標品で〔^<35>S〕GTP_γS結合活性をみたところ、いずれの膜標品においても活性上昇はみられなかった。したがって、シグマI受容体はG蛋白質と連関しない受容体であることが確認された。
