研究概要	岸本 1.IL-6のシグナルを負に制御する分子SSI-1およびSSI-1と相似性を有する複数の分子を同定し、これらの分子が種々のサイトカインのシグナルを負に調節していることを示した。 2.I型アレルギー疾患ではTh2細胞への優位な分化がその発症に中心的な役割を果すことを明らかにした。またアトピー性皮膚炎患者の単核球からは、IL-6やPGE2が過剰に分泌されており、それらがI型アレルギーで認められる過剰なTh2分化への責任分子である可能性を示した。 3.無ガンマグロブリン血症(XLA)の責任遺伝子であるBtkと相互作用する新規分子Sabを同定した。またBtkの生理的基質としてWASP(Wiskott-Aldrich症候群の責任遺伝子)を同定した。一方、フローサイトメトリー法によるXLA患者、保因者診断法を確立した。 4.マウスプレB細胞刺激因子(PBSF)の受容体としてケモカインレセプターCXCR4を同定した。ヒトCXCR4はエイズウイルスの宿主細胞への感染に必須の受容体であるが、マウスCXCR4もHIVの宿主細胞への侵入を支持することを明らかにした。山西 1.ヘルペスウイルスの潜伏感染・再活性化が免疫系に与える影響を解明することを目的として、ヒトヘルペスウイルス6(HHV-6)の潜伏感染時に特異的に発現するmRNA及び再活性化時に特異的に発現するmRNAを同定し、潜伏感染・再活性化の機序の一部を明らかにした。 2.ヘルペスウイルスには、免疫細胞の走化性、接着などに関係するケモカイン及びそのレセプターに類似の分子がコードされている。HHV-6感染の免疫系への影響を明らかにするために、HHV-6上のケモカイン様蛋白U83とレセプター様蛋白U12の性質を検討し、生理活性を明らかにした。木下 1.GPIアンカー生合成の第1ステップの酵素が、PIGA,PIG-H,PIG-C,GPI-1の4成分複合体であること、第2ステップの酵素がPIG-Lタンパク質であることを明らかにした。 2.GPIアンカーをタンパク質に結合するトランスアミダーゼが、GPI8とGAA1の2成分を含む複合体であることを示し、このうちGAA1のC末端がGPIアンカー付加シグナル配列を認識するサブユニットであることを示した。 3.GPIアンカーのドリコールリン酸マンノース合成酵素において、哺乳動物では2種類の酵素(DPM1,DPM2)が必須であることを証明た。 4.トリパノソーマのPIG-Bホモログをクローニングし第3のマンノースの転移に働くことを示した。菊谷 1.CD40のリガンド(CD40L)の遺伝子の変異が原因で発症する高IgM血症の自己免疫現象の機序を解明するために、CD40欠損マウスのリンパ球をヌードマウスへ移入した。結果、自己抗体の産生、自己免疫性の胃炎や甲状腺炎が誘導され、CD40-CD40L相互作用が自己反応性CD4T細胞の制御においても機能していることが明らかになった。 1.ヒト野生型CD40を発現するトランスジェニックマウスでは、B細胞の出現は正常であったが、抗体産生が著しく阻害されており、CD40が形質細胞分化を抑制するシグナルを伝達している可能性が示唆された。更に、この抑制シグナルはCD40の細胞質内領域の膜遠位部より伝達されていた。清野 1.炎症性腸疾患IBDの発症には、Th2系の禁止クローンの出現が発病に強く関わること、禁止T細胞クローンは腸管局所において消化管常在細菌の刺激により増殖しIL-4を自発的に産出して自己や食餌抗原に対する異常なB細胞応答を誘導することを明らかにした。 2.腸管の上皮細胞表面に腸管上皮細胞間リンパ球の機能を特異的に調節制御する分子が存在することを示した。 3.鼻粘膜関連リンパ組織(NALT)とパイエル板との類似点を見い出し、NALTが鼻粘膜関連組織における粘膜免疫誘導組織であることを示した。また、抗原を多量に経鼻投与する事により鼻粘膜での抗原特異的免疫寛容を誘導し、鼻粘膜アレルギー治療に対する治療法の可能性を示唆した。
