研究概要

SSI-1(STAT-Induced STAT Inhibitor-1)は、1997年にわれわれが単離し、その機能を明らかにした分子である。SSI-1のin vitroでの機能は、サイトカイン刺激により誘導され、サイトカインシグナル伝達の主要な因子であるJAKの活性を制御することでサイトカインシグナル伝達を阻害することにある。われわれは、SSI-1のJAKおよびサイトカインに対する特異性を解明するために、SSI-1を胸腺で強制発現させたSSI-1Tgマウスを作成し解析を行った。その結果、SSI-1Tgマウスはγcレセプターの欠損マウスと同じフェノタイプを示すことが分かった。また、SSI-1Tgマウスの胸腺細胞をIFN-γ,IL-4,IL-6,IL-7などのサイトカインで刺激し、JAKの下流の分子であるSTATのリン酸化を解析したが、それぞれのサイトカイン刺激に対応する各々のSTATのリン酸化が抑制されていた。これらの結果により、SSI-1はサイトカインやJAKに対する特異性を持たないと考えられた。また、われわれが前回、報告したように、SSI-1KOマウスはリンパ球,心臓、肝臓などのSSI-1発現臓器が加齢とともにアポトーシスを起こす。今回、SSI-1KOマウスに認められるアポトーシス誘導メカニズムを解明するために、SSI-1KOマウスの胎児線維芽細胞やSSI-1を強制発現させた線維芽細胞株を用いて解析を行った。その結果、SSI-1はTNF-αが誘導するアポトーシスシグナルの抑制分子でもあることが明らかにされた。さらに、SSI-1はTNF-αの下流において細胞の生存に働くシグナル伝達分子の1つであるp38の活性化を促進させることにより、TNF-αが誘導するアポトーシスを抑制していることも明かとなった。これは、今までに知られているSSI-1のJAK-STATの抑制メカニズムだけでは説明できず、新たなSSI-1の機能の存在を示すものである。
サイトカインの情報をnegativeに調節する分子が発見され、そのin vivoにおける役割が明らかになりつつあるが、一方で、生体内にいてサイトカインの異常産生が自己免疫疾患や血液系腫瘍の発生にかかわる例が報告されている。我々はIL-6のリウマチ様関節炎発症における役割を明らかにするため、IL-6ノックアウトマウスを用いて、An tigen-induced ArthritisやCollagen-induced Arthritisが誘導出来るか否かを検討した。その結果、これらのArthritisの発症にIL-6の存在が必須であることが明らかになった。このような基礎的研究の上に立って、現在ヒト型抗IL-6R抗体のリウマチ治療への応用がPhase1,2studyとして行われているが、我々はCastleman病においても抗IL-6R抗体が著明な効果を発揮することを明らかにした。
我々はこれまで、ケモカインSDF-1/PBSFとその受容体CXCR4が胎児の生存、胎生期のBリンパ球の生成、骨髄での骨髄球系造血、心室中隔形成、血管形成に必須であることを明らかにしてきた。そこで次に、その血球細胞自身における役割、成体における役割を明らかにするために、正常マウスとSDF-1/PBSF欠損マウス、CXCR4欠損マウスの胎児肝臓血細胞を成体マウスに骨髄移植する実験を行った。SDF-1/PBSF欠損マウス細胞を移植されたキメラマウスでは、骨髄、胸腺、脾臓においてリンパ球、顆粒球が正常の胎児肝細胞を移植したときと同様数認められた。これに対して、CXCR4欠損マウス細胞を移植されたマウスの骨髄のドナー由来のBリンパ球、顆粒球、胸腺のT細胞いずれも減少していた。また、このマウスの骨髄のBリンパ球細胞数の低下は他の系列の細胞や、脾臓の成熟B細胞数の低下と比較して著しかった。したがって、SDF-1/PBSFやCXCR4は、骨髄でのBリンパ球初期分化に特異的な作用があると考えられる。以上の結果より、血球上のCXCR4が成体のリンパ球系、骨髄球系造血の再構成に必須であることが明らかとなった。
ヒト無ガンマグロブリン血症(XLA)の責任遺伝子であるチロシンキナーゼBtkの、Bリンパ球における機能を明らかにすることを目的として解析を進めた結果として、Btkの細胞内における基質としてWiskott-Aldrich症候群の責任遺伝子であるWASPを同定し、そのBtkによるリン酸化部位の同定によってBtkの基質特異性、すなはちBtkによってリン酸化されるチロシン残基の周辺のコンセンサス配列を明らかにした。また、Btkと相互作用する新規分子Sabを同定し、SabがBtk活性の抑制因子であることを明らかにした。さらに、Btkの主たる結合分子が新しいリンカー分子BLNKであることを解明し、抗原レセプター刺激後に生じる細胞内カルシウム動員におけるBtkの機能を明らかにした。すなわち、B細胞抗原レセプターの刺激後、Sykによってリン酸化されたBLNKがBtkのSH2ドメインと結合することがBtkによるホスホリパーゼCγの完全なリン酸化に必須であることを示した。一方では、Btkの発現をフローサイトメトリーによって解析する新しいXLAの診断法を確立し、従来の分類不能型の免疫不全症とされていた症例のうち、かなりの数は実際にはXLAであることを示した。

山西グループでは、ヘルペスウイルスの免疫系に与える影響の解明を目標としている。血球系細胞において持続・潜伏感染を生じる、カポジ肉腫関連ヘルペスウイルス(KSHV,HHV-8)、サイトメガロウイルス(HCMV)、ヒトヘルペスウイルス6(HHV-6)、ヒトヘルペスウイルス7(HHV-7)に関して以下の結果を得た。
i)HHV-6,HHV-7,KSHVの感染、増殖などに関するウイルス学的研究。
HHV-6の2つのvariant(A及びB)のうち、HHV-6variant Bの全塩基配列を明らかにした。この結果、先に全塩基配列の明らかになっていたHHV-6variant Aとの間で6個の遺伝子が変異していることが判明した。Variant A,variant B間には、突発性発疹の発病の有無など病原性に差が見られるが、この原因がここで判明した遺伝子変異にあることが示唆された。
KSHVの増殖において早期に出現する遺伝子(vIRF,K5,ORF50)の転写産物、蛋白の局在、トランス活性化について解析を行い、KSHVのウイルス増殖における初期現象の一部を明らかにした。
ii)HHV-6,HHV-7,KSHVの遺伝子にコードされるサイトカイン・ケモカイン類とその受容体に関する研究。
HHV-6遺伝子にコードされるケモカイン遺伝子U83及びケモカイン受容体遺伝子U12を同定し、これらが細胞の走化性などに関係していることを示唆した。ケモカイン及びケモカイン受容体は、ヒト免疫不全ウイルス(HIV)の細胞側受容体として注目されている。HIVと同じくCD4陽性T細胞に感染するHHV-6がこのような遺伝子を持つことは、生体内における両ウイルスの相互作用を示唆している。
また、KSHVは、その遺伝子上に多くの宿主遺伝子を取り込んでいることを特徴とする。その1つである、ウイルス性のインターロイキン6(IL-6)が、ヒトのIL-6の産生を誘導することを示した。これは、KSHVとの関係が指摘されている、キャッスルマン氏病の病因を示唆するものである。
iii)HCMV,HHV-6,HHV-7の潜伏感染・再活性化の機序およびその生態に与える影響の研究。
HHV-6Bの潜伏感染時に発現するmRNAを見出した。次に、HHV-6Bと近縁であるHHV-7の潜伏感染遺伝子を同定した後、HHV-6Bのものと比較し、両者においてアミノ酸配列が保存されている蛋白を見出した。これらの蛋白は、血球系細胞のシグナル伝達物質Cyclophilin Bリガンド(CAML)の機能を抑制することが、Yeast Two-hybrid法などにより見出されたが、この性質はサイトメガロウイルスの潜伏感染蛋白にも保存されていた。このことは、これらの蛋白が、ヘルペスウイルスの潜伏感染にとって、進化的に重要であることを示している。これらは、ウイルス再活性化シグナルの伝達を抑制することより、潜伏感染を安定化すると考えられた。

I)宿主側の20数個あると予想されるGPIアンカー生合成遺伝子群に関し進展があり、その結果16遺伝子が明らかになった.
(1)ドリコールリン酸マンノース合成酵素の新規成分DPM3をクローニングし、活性中心を持つDPM1をこれが正に制御すること、DPM2はDPM3を介して間接的にDPM1を正に制御することを明らかにした。
(2)第1のマンノースを付加するPIG-Mが小胞体の内腔側で働くことを示し、GPI中間体が細胞質側から内腔側へフリップするのはこのステップの直前であることを明らかにした。
(3)第1のマンノースにエタノールアミンリン酸を付加するPIG-Nをクローニングし、その酵母ホモログがGPIを阻害する抗真菌剤であるYW3548の標的であることを示した。
(4)タンパク質をつなぐエタノールアミンリン酸を、第3のマンノースに付加するPIG-Oをクローニングし.これがPIG-Fとの複合体として機能することを証明した。
II)病原微生物のGPI生合成に関し、トリパノソーマと熱帯熱マラリア原虫のPIG-Mをクローニングした。
III)発作性夜間血色素尿症の異常クローンは、8年にわたって造血を支配し得ることを示した。

消化管上皮細胞層における胸腺外分化T細胞ならびにCMISに依存しないB-1細胞の発達と分化機構を解明するために、T3bプロモーターを用いて腸管局所にIL-15を強発現したトランスジェニックマウスを作製した。
T細胞レセプターα鎖欠損マウスに発症する炎症性腸疾患の引き金としてTh2型にバイアスしたβB1細胞が関与すること。またこのバイアスにはBacteroides属細菌が重要なことを明らかにした。
さらにIL-4モノクローナル抗体の接種が病態の制御に有効であることを証明した。
IL-7R KO,alyfaly,LTaKOマウスを用いて、鼻咽頭関連リンパ組織の組織構築は、消化管関連リンパ組織のそれとはまったく異なるカスケードを介していることを明らかにした。
消化管の粘膜固有層にはIL-5R陽性、IL-15R陽性のB-1B細胞が多数存在すること、またIL-15を投与するとこの腸管B-1細胞のIgA産生能が著しく亢進することが明らかになった。
粘膜系Th2型細胞による食餌アレルギーのモデルマウスを作製した。
上皮細胞と粘膜系リンパ球のトランスウエル混合培養システムを用いると、粘膜免疫応答を開始する端諸であるUEA-1陽性のM細胞様のユニークな細胞を誘導できることを明らかにした。

1.EBウイルスの形質転換因子LMP1がCD40シグナルと同様にクラススイッチを含むリンパ濾胞外でのB細胞活性化と分化を誘導するが、一方でCD40とは異なり胚中心B細胞分化を抑制することを示し、LMP1がCD40のシグナルの一部を利用してEBウイルスの初期感染に寄与している事を明らかにした。
2.リンパ球セマフォリンCD100による刺激はCD40で誘導される増殖や免疫グロブリンのクラススイッチを増強すると共に、in vivoでも抗体産生を増強する事を示した。さらに、CDl00の機能的受容体がCD72である事を明らかにした。
3.CD100欠損マウスやCD100トランスジェニックマウスを作製し、CD100が、CD5陽性のB1細胞の分化、CD40YLPSに対するB細胞の反応性、T細胞依存性抗原に対する抗体産生反応に必要である事を示した。
4.ペプチドライプラリーを用いて、NOD由来膵島反応性T細胞クローンが認識するペプチドの配列を決定した。更に、このペプチドをNODマウスに投与すると自己免疫性糖尿病の発症が抑制される事を示した。
