研究概要	岸本:1)JAKの制御蛋白として同定されたSOCS-1(Suppressor of Cytokine Signaling-1)が、in vivoで、LPSシグナルにより、誘導されLPSシグナルを阻害することや、LPSトレランスおよびLPS/CpG, LPS/MALP2等のクロストレランスにも関与していることを、SOCS-1欠損マウスを用いて解析し明らかにした。また、このLPSシグナル阻害は、STAT1を介さず直接TLRシグナルに作用することもSOCS-1/STAT1 2重欠損マウスを用いて明らかにした。 2)SOCS-1がTh1,Th2分化の制御に重要な作用をもつ分子である事を、SOCS-1ヘテロ欠損マウスの解析により明らかにした。山西:ヒトヘルペスウイルス(HHV)-6および8に関して: 1.HHV-6の潜伏感染特異的遺伝子および蛋白を同定し、再活性化の起点となるウイルス遺伝子を同定した。また、HHV-6の潜伏感染において、他のヘルペスウイルスで知られていなかった、新しい相の存在を明らかにした。 2.HHV-6感染の細胞側の受容体として知られているCD46の、HHV-6感染に関わるドメインおよび、これに結合するウイルス側の蛋白を明らかにした。 3.HHV-8の再活性化に重要なことが知られている、ウイルス蛋白RTAによるウイルス遺伝子K9の発現を解析し、既知のものとは異なったcis-acting elementを見いだした。木下:1.GPIアンカー型タンパク質に結合して細胞を溶解する細菌毒素であるエロリジンが、GPIアンカーとN結合型糖鎖の両方を認識して高親和性結合することを証明した上で、エロリジンを用いたGPIアンカー欠損細胞株の樹立法を確立した。 2.ヒトのGPIアンカー生合成遺伝子群に関し、新たに樹立したGPI欠損細胞株を用いることにより、新規の3遺伝子をクローニングし、うち一つ(PIG-U)の機能を明らかにした。 3.発作性夜間血色素尿症の発症機構に関し、GPIアンカー欠損造血幹細胞は細胞傷害性リンパ球に抵抗性で、正常幹細胞を凌駕して拡大し得ることをマウスモデルで示した。これにより、再生不良性貧血患者においてしばしばGPI欠損クローンが拡大するメカニズムが示された。菊谷:樹状細胞上に発現しT細胞刺激活性を有するセマフォリンSema4Aの受容体の同定を行った。リコンビナント可溶性Sema4Aの種々の細胞への結合を指標に、Sema4A受容体が活性化T細胞と一部のT細胞株(EL4等)上に発現することを明らかにした。次に、EL4細胞から発現クローニング法で、Sema4A受容体としてTIM(T cell and immunoglobulin-like domain and mucin domain)ファミリーのメンバーTIM-2をクローニングした。また、Sema4A刺激により、TIM-2の細胞質内領域にチロシンリン酸化が誘導されことから、TIM-2はSema4Aのシグナルを細胞内へ伝達していることが示唆された。審良:TLRを介したシグナルでは、TIRドメインを持つアダプターMyD88が全てのTLRを介した炎症性サイトカインの産生に必須である。しかし、TLRごとに遺伝子発現の誘導が微妙に異なることが明らかになってきた。今年度、TIRドメインを持つもう一つのアダプターTIRAPの生理機能を、ノックアウトマウスの作製により解析した。その結果、TIRAPがTLR2,TLR4のMyD88依存性のシグナル伝達に必須であることを明らかにした。さらにTIRドメインを持つ3番目のアダプターTRIFを同定し、TRIFがMyD88やTIRAPと異なりIFN-βプロモーターを活性化させることを明らかにした。TIRドメインを持つアダプターが、TLRを介したシグナルの特異性を規定していることが示唆される。
