陽イオンポンプで見出した順次的ATP結合と多重リン酸化;オリゴマー構造と機能調節

研究概要

1価陽イオン輸送ATPasesは、ATPによりリン酸化反応中間体(EP)を形成し、P型ATPaseと称される。例えばNa/K-ATPaseは、Na^+に依存したEP形成と共役しNa^+を能動輸送、ついでK^+に依存したEP分解と共役してK^+を能動輸送すると、生化学、分子生物学の教科書に記載されている。しかし我々はNa/K-ATPaseが4量体、(αβ)_4、構造をとり、EPは単にそのうちの半分、(αβ)_2、でのみ形成され、最初に2つの(αβ)_2でそれぞれE1Pが、そして残りの2つの(αβ)_2にそれぞれATPが結合する、ついでE1PはE2Pに変化、E2PがK^+で脱リン酸化されると、脱リン酸化された2つの(αβ)_2にそれぞれ2分子のK^+が閉塞され、他の2つの(αβ)_2にATPが共に結合していることを見出した。Na/K-ATPaseに結合しているEATPに関しては非常に外部のATPと交換しやすい明らかにされた。一方、4量体構造を可視化する試みを行い、FITCラベルNa/K-ATPaseのFITC蛍光が量子化された状態で消光することを示唆する結果を得た。
H/K-ATPaseでは同様に、H^+-ATPase反応中半分の(αβ)にEPが、残りの(αβ)にATPが結合していることを見出した。この場合低濃度のATP存在下ではATP結合は存在せずEPのみが存在する。高濃度ATP存在下ではEP:EATPが共存し、EATPは次の反応サイクルでEPを経由して分解されることも示された。
