研究概要	Shewanella putrefaciensはさまざまな重金属を電子受容体として生活できる特異なバクテリアである。92残基のアミノ酸からなる4ヘムタンパク質(チトクロムc_3)はこのバクテリアが持つ電子伝達タンパク質の一つである。これは現在までに報告されているものでは最も分子量の小さい4へムタンパク質であり、へム密度は著しく高い。昨年度と今年度のK.Nealson教授らとの共同研究でその酸化還元的性質をかなり明らかにすることができた。3つのヘムには主たる還元ステップが一応あるといえるが、1つのヘムには特に大きな還元ステップが見られない。相互作用ポテンシャルも正の値、負の値ともに著しく大きなものがあった。また、c型チトクロムを多く持つこのShewanella putrefaciensを宿主にして硫酸還元菌Desulfovibrio vulgaris Miyazaki F株のチトクロムc_3(107残基)遺伝子の発現系をつくることに成功した。今年度は平成11年6月にウィスコンシン大学のFillingame教授が横浜国立大学を訪れてエネルギー変換系タンパク質の遺伝子発現系について討論を行った。平成12年1月には研究代表者阿久津が渡米して、カリフォルニア工科大学にてNealson教授、Tsapin研究員と共同研究のまとめかたについて討論を行い、さらにマサチューセッツ工科大学にてGriffin教授と固体NMRの利用について討論を行った。平成12年3月に研究分担者の小澤潔助手が渡米してカリフォルニア工科大学でNealson教授と、アリゾナ大学でCusanovich教授と実験データについて討論した。さらに、その後、カリフォルニア工科大学のTsapin研究員が来日し、横浜国立大学において研究代表者、分担者とともに研究のまとめを行った。
