研究概要	本研究において、品川・岩崎のグループは大腸菌・古細菌・酵母を研究材料にして、二重鎖切断の組換え修復の機構の研究を行い、海外の大腸菌の組換え、古細菌の組換え、酵母の組換えの研究のエキスパートのグループと交流して共同研究を進めた。品川・岩崎らは大腸菌の組換え修復の後期過程に主要な役割を果たすRuvA,RuvB,RuvCのミュータントタンパク質を多数作成して、機能に重要なアミノ酸を同定し、反応機構の解析を行った。古細菌においては組換え中期過程に重要なRecAタンパク質のホモログであるRadAタンパク質が機能的にもRecAと同様な働きをもつことを明らかにした。分裂酵母で組換え修復に関与する遺伝子群を同定し(rhp57)これが出芽酵母のRad57の機能的・構造的ホモログであることを証明した。 A.M.Carrらは分裂酵母で、プロテインキナーゼRad3はDNA損傷とDNA複製の停止の両方に応答して細胞分裂を停止させるように働くことを明らかにした。 A.J.Clarkらは種々の古細菌からPCRでradA遺伝子を単離し、系統樹を作成し16SRNAによる分類と比較した。 M.M.CoxらはRecAタンパク質による相同的DNA組換え機構を解析し、RecAはDNA鎖交換反応において、二本鎖同士の対合の中間体の形成を行わせるのではなく、RecAのモータータンパクとしてのATPに依存した二本鎖DNAのゆがみがおこさせることが必要であることを証明した。
