研究概要

Plasminogen activator(PA)と、PAの阻害タンパク質であるtype-1 PA inhibitor(PAI-1)は、PA-plasmjlcascadeのtrigger′どPA活性の生理的調節物質どじて重要な因子である;ﾟ木研究でほ:ﾞPA-plasmin cascadeの調節機構について主にPAI-1の遺伝子発現の調節機構について追究し、以下の知見を得た。
1) マウス肝細胞株AML12を用いてPAI-1遺伝子発現のサイトカインによる調節調節について検討しIL-1、TNFαがPAI-1mRNA発現を誘導することを明らかにした。IL-1はPAI-1mRNAの安定性にはほとんど影響を及ぼさず転写を顕著に活性化した。
2) IL-1β遺伝子欠損マウスの急性期モデルにおけるPAI-1遺伝子発現の特徴について検討を行い、テルペン油による局所炎症モデルにおけるPAI-1誘導にIL-1がメディエーターとして重要な役割を果たしている可能性が示唆された。
3) PAI-1遺伝子の転写後調節機構について検討しサイクリックヌクレオチドにより調節を受けるcis-エレメントを同定した。またTGFβによるPAI-1転写誘導機構についで検討しsmedの役割を明らかにした。
4) PAI-1遺伝子欠損マウスを用いて、PAI-1の異なるエピトープを認織するモノクローナル抗体の作成に成功しELISAを構築した。
