研究概要

T細胞抗原レセプターを介するT細胞の活性化におけるCD3分子の細胞内領域にあるチロシンモチーフ(ITAM)の役割について解析を行った。CD3ζ鎖のITAMを変化させたζ鎖を発現するトランスジェニック(Tg)マウスを作製し、ζ鎖欠損マウスと交配することによってζのITAMのチロシンのみに変異を有するマウスを樹立した。このマウスを更に,TCR-Tgマウスと掛け合わせたマウスのT細胞を解析した結果、活性化の状況、機能発現、細胞分化において殆ど大きな差違はなかった。この結果は、ζ鎖のITAMはredundancyがあり、他のCD3分子のITAMによって担われていると考えられる。一方では、ζ鎖のリン酸化の程度とT細胞のアナジーとの関係が報告されており、その関係を明らかにしなければならない。ζ以外のモジュールを介するシグナルの伝達が重要であり、ζには結合せずCD3εと結合する唯一の分子としてのCASTは重要であると思われた。細胞内輸送シグナルとしてのチロシンシグナルの解析としては、今回新たにAP1複合体中のμlに相同性を有するμ1Bをクローニングし、そのゲノム構造も明らかにした。μ1Aがubiquitousna発現なのに対して、μ1Bは小腸や膵臓などの極性を有する細胞に高発現しており、極性を持った細胞内輸送に関係するAP複合体を形成すると考えられた。一方、この間存在が明らかになったAP3複合体は、AP1,AP2と同様にクラスリンと会合してクラスリン被膜小胞を形成して蛋白輸送を司ることが明らかになった。
