研究概要	RNAポリメラーゼIIによるRNA鎖伸長反応を制御する因子として同定されたDSIF(DRB sensitivity-inducing factor)は、転写阻害剤DRBやH-8の転写阻害に必須な転写因子である。DSIFは、DSIFp160とDSIFp14の2つのサブユニットから構成され、p160とp14はそれぞれ酵母Spt5とSpt4のヒト相同性因子である。DSIFは、RNAポリメラーゼIIに結合してRNA鎖伸長反応を制御する。我々の生化学的解析から、DSIFには、転写伸長反応を負に制御する場合と正に制御する活性が存在し、負の制御には、新規の転写伸長因子NELF(negative elongation factor)の関与が示された。さらに、DSIFとNELFによる転写抑制は、P-TEFb(positive elongation factor b)により解除され、その制御はRNAポリメラーゼIIの最大サブユニットのC末端ドメイン(CTD)のリン酸化と密接に関連していることが明らかとなっていた。今年度の研究は、高度に純化されたタンパク質による試験管内転写再構成系を用い、DSIF、NELFおよびP-TEFbによる転写伸長反応制御機構を検討した。その結果、クロマチン構造を有した鋳型DNAからの転写伸長反応に関与する因子FACT(facilitate chromatin transcription)が、P-TEFbと協調的に働いてDSIF/NELFによる転写阻害を解除することを見出した。
