研究概要

本研究は、第一級の国際的な研究者による共同研究のもとに、細胞周期を統御する細胞内シグナル伝達系を、多様な変異遺伝子を用いて解析する事を目的とする。近年の研究から、細胞の増殖は、チロシンリン酸化を介するシグナル伝達系による正の制御と細胞周期制御因子による負の制御からなるシステムよる統御をいけている事が明らかになっている。そこで、本年は、チロシンフォスファターゼSHP-2の結合蛋白質として同定され、まだ十分その機能が明らかになっていないSHPS-1について解析をすすめた。その結果、v-Srcによる癌化は、SHPS-1の発現を抑制する事、SHPS-1の遺伝子をクローニングし、v-Src癌化細胞にに導入する、癌細胞の増殖を選択的に阻害する事が明らかになった。
