種々の病態における新しいカルシウムシグナリングのゲノム薬理学的研究

研究概要

カルシウム結合蛋白質が、Parkinson病、Alzheimer病などの中枢神経障害、心筋梗塞などの虚血性疾患や悪性腫瘍など種々の疾患において発現変化が認められ、その病態における役割が注目されている。我々は前年度本研究において、カルシウム結合蛋白質研究の欧州会長であるC.W.Heizmannらと共同研究し、我々が独自に見出したカルシウム結合蛋白質、S100Cが種々の疾患における発現変化などを見出した。今回、カルシウム結合蛋白質研究の若き世界的リーダーであるBerchtoldM.W.らも加え、S100Cを中心とした新しいカルシウムシグナリングと病態における役割やその分子機構について検討した。さらにS100ファミリーの組織分布、細胞内局在、細胞周期依存性の局在変化における細胞骨格との関連性を検討した。その結果、S100ファミリーに属する蛋白質は、構造が非常に似ているにも関わらず、細胞や組織における発現、細胞周期や種々の刺激による細胞内再局在が非常に多面的であることが明らかになった。特に、S100A1、S100A4、S100Cは細胞質に存在し、筋小胞体やストレスファイバーに結合して認められたのに対し、S100A2は細胞核にのみ存在した。また、Ca^<2+>、Zn^<2+>、Cu^<2+>などの金属イオンに対する親和性においてもその細胞内機能および細胞外機能と関わる特異性があることが共同研究者であるC.W.Heizmannとの共同研究で明らかとなった。各々の、標的蛋白質についても、多くの蛋白質が同定され、現在その生埋的磯能について検討中である。さらに、S100Cは細胞内カルシウム濃度によりその遺伝子発現量か調節されている可能性が示唆されたことや、肺癌、大腸癌においてその遺伝子が増加している結果が得られているが更に詳細に検討し、遺伝子発現制御機構などに焦点をあて現在解析中である。
