HIV感染を阻害する生理活性ペプチドの創生

研究概要	HlVの逆転写酵素(RT)のアミノ酸置換により活性が消失するホットスポットの部分に対するアンチセンスペプチドを合成し、McGill AIDSセンターのKleiman博士のもとに送り解析したところ、RT活性を強力に抑制した。しかし、このペプチドは細胞内には入りにくいので、そのアミノ酸配列をコードするcDNAを合成し、これをプラスミドに組み込んだものを作成した。 HIVのgp120やgp41の分子内アンチセンスホモロジーボックス(AHB)やHIV感染の受容体の一部として働くケモカインレセプターであるCXCR4やCCR5のAHBの解析を行った。それらのAHBのヘキサマーペプチドを150種類ほど合成し、HIV感染阻止効果の検討を行った。HIV感染阻害は、MT4細胞にHIV-IIIBを感染させ、感染による細胞死をMTTT法で検出するときの細胞死の阻害により検出した。阻害効果を持つペプチドが数種類得られたが、治療薬につながるような強力な抑制作用を持つものを得るには至っていない。ペプチド鎖をもう少し長くする必要があると考えられる。一方、T22ペプチドとしてgp41の分子内ペプチドが強力な感染阻害効果を持つことが報告された。T22は30アミノ酸から成る比較的長鎖のペプチドであるが、gp41のAHBに一致することが我々のAHB解析でわかったので、AHBペプチドは長鎖のものを合成する必要があると考えられた。 Monash大学のCosic博士の電磁気共鳴解析法によりT22の解析を行ったところ、CXCR4と電磁気共鳴波が一致していることがわかった。従って、AHB法と電磁気共鳴解析法の両方で一致して予測される部分のペプチドが、有効な活性を発揮するペプチドの候補として検討すれば検索効率を上げることができると考えられる。フランスのKazatchkine博士には、HIV感染患者の血漿等の解析用試料での協力をお願いした。
