生物進化における遺伝暗号の変化に対応する翻訳終結因子の役割に関する研究

研究概要

遺伝暗号はあらゆる生物に共通・普遍のものであると信じられてきた。しかしわれわれのテトラヒメナにおける翻訳終止コドン異常使用の発見を契機に、遺伝暗号といえども固定化されたものではなく、生物進化にともなって変化するものであるということが明らかになってきた。しかし遺伝暗号の使用パターンに変化をもたらす原因因子が何であるかについては現在のところ全く不明である。遺伝暗号の認識には多くの因子が関わっているが今回は特にポリペプチド鎖終結因子(RF)に着目し、翻訳終止コドンUAGおよびUAAをグルタミンのコドンとして使用するテトラヒメナ、UGAコドンをシステインのコドンとして使用するEuplotesのRFの質的変化の有無を調べた。これまでに報告されている種々の生物のRFでとくに高度に保存されているアミノ酸配列をもとに、テトラヒメナおよびEuplotesでの遺伝暗号使用パターンを参考にしてPCRプライマーを作成し、両生物の全染色体DNAを鋳型としてPCR反応を行った。増幅されたDNAの構造解析からRF因子をコードする遺伝子の一部が増幅されたと確認できたものを選び出し、それらをプローブとして用いることによって完全長の両生物のRF遺伝子の単離・クローニングを行った。得られた遺伝子の構造解析の結果、予期に反してテトラヒメナおよびEuplotesのRF因子の遺伝暗号認識に関わる領域、とくにペプチドアンチコドンと呼ばれる領域においてこれまで報告されている酵母やほ乳動物のものとの間で大きな違いが認められなかった。そこで現在Euplotesにおいては通常のシステインのコドンUGUおよびUGCを認識するtRNAおよびそのtRNAのアミノ酸受容に関わる酵素の検出、テトラヒメナにおいてはUAGおよびUAAをグルタミンのコドンとして認識するtRNAのアミノ酸受容に関わる酵素の検出を試み、それら因子の構造変化と翻訳終止コドン異常使用との相関性について検討している。
