グロビン遺伝子群の多型を用いた日本人の起源に関する分子人類遺伝学的研究

研究概要	βグロビン遺伝子内のサラセミア変異とβグロビン遺伝子群内の多型とに注目し、日本人の起源を検討した.なかでも北方起源の可能性を見るために韓国、台湾の、また南方起源の可能性を見るためにタイ、マレーシア、インドネシア、モルジブのサラセミアの変異を決定し、変異に連鎖したグロビン遺伝子群の複数の多型の組み合わせからなるパターン(ハプロタイプ)と、遺伝子内の多型の組み合わせ(フレームワーク,FW)を解析した. 日本人に見られる8種類の変異が韓国人、中国人、タイ人、マレーシア人、インドネシア人、モルジブ人いずれかにも見られことが分かり、この変異に連鎖しているハプロタイプとフレームワークを解析した.それによるとcodons41/42 4bp欠失、codon17(A→T)、IVS-2 nt654(C-T)、initiation codon(T→G)は中国人由来と考えられた.日本人と韓国人とのみで共通の変異、つまりIVS-1 nt130(G→C)、IVS-2 nt1(G→A)、codon121(G→T)に関しては日本人由来か韓国人由来かは同定できなかった.興味ある変異はcodon15(G→A)である.これは東南アジア人とのみ共有していた.このハプロタイプの解析を行ったところ、日本人とタイ、インドネシア人とではハプロタイプやフレームワークとも異なっており、codon15(G→A)の起源は日本人と東南アジア人とでは異なっていると結論された. 以上から、日本人の起源をサラセミア変異および多型から検討した結果、中国・韓国人起源が主であり、東南アジア人起源の可能性は低いものと考えられた.今後さらに東南アジアでの日本人と共有する変異の検出とハプロタイプの解析が必要であると考えられる.
