カリウムイオンによるDNAの構造遷移を利用したリボザイム活性のスイッチング

研究概要

鉛リボザイムにおける金属イオン(鉛イオン及びマグネシウムイオン)の結合部位をNMRにより同定した。結合部位は2カ所あり、この各々に2つの金属イオンが競争的に結合する事がわかった。そしてこの知見に基ずいて、様々な金属イオンの組み合わせによる、切断活性の増進及び抑制を統一的に理解できるモデルを提出した。
又DNA結合性の抗ガン剤VCH9とDNAの複合体の構造決定、RNA結合性の蛋白質hmRNPDφ及びMusashi1の構造決定も行い、核酸構造の遷移と生物学的活性との相関について明らかにした。
