硫酸化多糖による蛇毒中和効果の証明

研究概要

硫酸カードランによる補体活性の阻止効果
犬血清中のC3を活性の指標として、放射一元免疫拡散法によって判定した。
補体の活性剤としてはキトサンおよびガラガラヘビ毒(Crotalus V.Viridis,Sigma,USA)を10mg/mlの濃度で用いた.硫酸カードラン(味の素、東京)の分子量は79+/-0.6kDaで、硫酸化度は15.2+/-0.3%である。硫酸カードランはキトサンおよびガラガラヘビ毒による補体の活性を完全にブロックした.
蛇毒による皮下出血の阻止効果
出血阻止実験は兎を用い、毒の投与量を0.02mgとしてCRDSを0.01,0.02,0.04,0.08,0.16,0.32mgまで変化させた.その結果、CRDSの量が蛇毒の1/2および等量の場合でも明らかな出血阻止効果が認められた。CRDSが2倍以上の例では完全な出血の阻止効果が認められた。さらに、実際面での有用性を明らかにするために0.02mgの毒を投与した後に、同部位に時間をおいてCRDSを投与し、その効果を検討した。皮膚区分は頭側の2カ所が毒のみ、尾側の2カ所をCRDSのみのコントロールとした。安楽死を毒投与後4時間目に実施し、出血の有無を確認した。その結果、30分後および1時間後にCRDSを投与した兎No.5では出血は認められるものの、コントロールと比較して明瞭な阻止効果が認められた。
以上の検討によってカードラン硫酸はキトサンおよび蛇毒による補体の活性作用をブロックする機能があることを証明し、ヘビの咬傷の治療薬として極めて期待できる物質であることを明らかにした。
