標的認識性糖鎖を配置した高分子ミセルの構築とその薬物ターゲティングへの展開

研究概要	前年度において、片末端に位置特異的に糖鎖を結合させたブロック共重合体(sugar-PEG-PLA)が得られることを明かとした。この成果をふまえて今年度は、末端に結合する糖鎖の種類を拡張するとともに、ブロック共重合体の自己会合に基づくcore-shell型高分子ミセルの調製法の確立とその特性解析を詳細に行った。ブロック共重合体を良溶媒であるDMAcに溶解させ、PEG鎖の選択溶媒である水に対して透析することによってPLAを内核、sugar-PEGを外殻とするsugar-micelleを調製した。このミセルは、動的光散乱測定結果から粒径約40nmの単分散ミセルであることが確認された。キュムラント解析による多分散度は0.07-0.09と極めて狭く、また、拡散係数の角度依存もないことから、ほぼ球形の形態を取ることが示唆された。高分子ミセルを薬物のキャリアとして用いる場合には、高度希釈条件における安定性が要求される。これは臨界会合濃度(cac)から見積もることが可能である。cacについては、蛍光法により決定した。その結果、cacは数mg/Lと極めて小さい値を取り、このミセル系が希釈に対して安定であることが明かとなった。次に、内核の運動性を見積もる目的で高分子ミセルの^1H-NMRを重水中で温度を変えて測定し、PLAとPEG由来のピークの積分比を算出した。PLAのT_gはおよそ40℃であるが、その付近の温度を境にしたPLA主鎖の運動性変化とピーク強度比の値は良く対応し、ミセル内核が温度上昇に伴って40℃付近でガラス状からゴム状へと転移することが示唆された。この事実は、内包薬物の放出を温度によって制御できる可能性を示しており、ガン温熱療法との沸用への展開が期待される。
