研究概要

本研究では、生体分子を脂質膜へ効率的にディスプレイするための技術の開発を行い、また、ドラッグデリバリー、バイオセンシングなどのナノスケールでのターゲッティングヘの応用を目的としている。本年度・研究代表者である松永は、有用タンパク質-MagAアンカー複合体(生体分子アーキテクチャー)の磁性細菌粒子上への高発現系の確立を目的とし、高発現ベクターの構築を行った。また、膜貫通領域を2つまで断片化したMagAアンカー分子のディスプレイに成功した。さらに、磁性細菌粒子上での分子アーキテクチャーの挿入条件を検討し、in vitroにおいてもMagAがアンカー分子として利用できることを確認した。また、研究分担者である小宮山は、モレキュラーインプリント法を用い、シクロデキストリンの架橋反応を解析し、二級水酸基側が架橋されることを見出した。また、ジペプチドPhe-Phe(D体)・抗生物質Cefazolin及びVancomycinを水溶液中で認識する人工レセプターを合成した。後藤は、W/Oエマルジョンを用いる複合化酵素の調製法が、有機溶媒中で高活性を発現させる新手法として有効であることを確認し、様々な糖分子へのアルキル鎖の導入法として利用した。酵素としてスプチリンを界面活性剤で複合化することによって、単糖類、二糖類へ高い転下率で長鎖アルキル基を導入できた。合成したラクトースのアルキル誘導体(LacCn)をハイブリッドリポソーム(DMPC/Tween20)に組み込み、in vitroにおける種々のガン細胞に対する制ガン効果をWST-Assay法によって検討した(A04班上岡教授との共同研究)結果、糖脂質を組み込んだリポソームが、脳腫瘍、肺ガン、肝臓ガン等様々なガン細胞の増殖を特異的に制御することを確認した。小畠は、プロテインAのBドメイン遺伝子をタンデムに5つ連結したB5DNAとシグナル配列を含むN末端領域のペプチドをコードする遺伝子配列に連結して、脂質修飾抗体結合タンパク質の発現プラスミドを構築した。大腸菌による遺伝子発現を行ったところ、B5タンパク質は脂質修飾されていることが分かった。また脂質部位をアンカーとして、リポソーム表面に配向固定できることが示された。得られたリポソームに抗体分子を結合し、抗原認識機能を有するイムノリポソームを作製できた。
