研究概要

より高度な機能を持つターゲティング原理や新規ターゲティング素子材料の開発とダブルターゲティング能を有する分子システムの工学的構築について下記の研究を行った。
(l) 小林は、抗体・薬剤コンジュゲイトや薬剤封入リポソームを作成し、ターゲティング機能をもったミサイル薬剤の創製を試みた。熱や超音波などの物理的刺激によって必要な時にのみ薬物を放出(時間的ターゲティング)できるようにし、高い標的指向性のあるモノクローナル抗体によって標的部位に製剤を集め、その部位にのみ薬物を放出(空間的ターゲティング)する、ダブルターゲティング薬剤も開発した。
(2) 岡野はこのような高度な薬物治療法をさらに発展させるために、疾病に伴って生体から発せられる信号を感知して初めて薬物を放出し、その信号が消失したら薬物放出が止まるといったインテリジェントシステムの構築について研究した。
(3) 飯島は生体高分子による細胞ターゲティングを検討し、シアル酸を基本とする転移阻害性ポリマーによるガン転移の阻止を目指した。このため、腫瘍細胞の表面分子に類似した、内皮細胞に特異的に認識されるターゲティングポリマーの開発を試み、よい結果を得た。
(4) 関は植物培養細胞系の多様性を評価し、特定の性質を有する細胞をターゲティングして系から分離する手法を開発し、抗ガン剤として有望なタキソールなどの二次代謝産物高生産株の選抜に応用してよい結果を得た。
(5) 末永は、生体が発する微小信号を検出するために、組織レベルで情報を拾える超マイクロ電極型デバイスを開発した。そして、細胞近傍の酸素濃度をマッピングすることにより、ヒト結腸腺ガン細胞の呼吸活性、ハネモプロトプラストの呼吸および光合成活性のイメージングを、単一細胞レベルで行うことに成功した。また、呼吸を阻害するCN-などの化学物質を添加した際の細胞イメージ変化を、シミュレーション法を用いて解析することにより,化学物質の細胞膜透過性を評価することができた。
