研究概要

生体内の重金属を結合するSH基高含有タンパク質をタンパク質工学的にデザインし直し、アミノ酸配列を人工的に改変して、特定の重金属を識別する金属タンパク質を創製し、重金属解毒や医療医薬または環境中の重金属モニターや集積除去に関わる新素材機能の開発を目的とし、ヒト由来の重金属結合タンパク質メタロチオネインを立体構造と金属結合機能解析のモデルに下記の研究を行った。
1. 大腸菌による重金属結合タンパク質の生産系の開発:
GST-hMT-II融合法によって、ヒトメタロチオネインおよびその変異タンパク質を大腸菌で大量発現させることができた。
2. 重金属結合タンパク質の発現生産効率評価のための簡易測定法の開発:
大腸菌で大量生産したメタロチオネインタンパク質の可溶化条件を検討し、可溶化の後、FactorXで切断して、メタロチオネインタンパク質の精製条件を確立した。現在、米国ハーバードメディカルスクールにおいて、野生と変異タンパク質の生化学的諸性質を解析中である。
3. 三次構造モデルの構築と重金属特異的結合能を有するタンパク質の開発:
三次構造モデルを構築し、コンピューターでディスプレイできるソフトを開発した。
