BおよびC型肝炎ウイルス受容体の解明と感染制御

研究概要

ヒトB型肝炎ウイルス小型球形粒子を用いてヒト肝におけるウイルス受容体を調べた。以下に結果の要点を述べる。
1) ヒト肝臓より抽出した糖タンパク質画分(human Hepatic glycoprotein:hHGP)に、ヒト肝がん細胞由来huGK-14細胞より産生される非感染症B型肝炎ウイルス小型球形粒子(以下これをHBsAgとする)が特異的に結合することを見い出した。この結果から、今回見いだされたhHGPはヒトB型肝炎ウイルス小型球形粒子の受容体として機能している可能性が示された。
2) このHBsAgのhHGPに対する結合には、hHGP中に存在するシアロ糖鎖が必須であることを見いだした。シアロ糖鎖の性状を種々のレクチンを用いて調べた結果、Neu5Ac2-6Gal結合を持つ糖鎖が存在する可能性を見いだした。シアリダーゼ処理により結合活性が消失することからhHGP中のシアル酸残基が結合に重要であることが示唆された。
3) hHGPの部分精製に成功した。さらに、HbsAgは、肝臓組織の他、ヒト膵臓組織に結合性を示すことを確認した。
4) ヒトB型肝炎ウイルス小型球形粒子は、ヒト膵臓がん細胞のムチン画分にも結合性を示すことを見いだした。以上の結果から膵臓組織にもHBsAg受容体が存在する可能性が示唆された。 ヒト膵臓がん細胞のムチン画分の部分精製に成功した。
5) ヒト肝およびヒト膵臓がん細胞から得た糖タンパク質およびムチン画分に対するモノクローナル抗体の作成を達成した。これにより、HBV受容体の組織間分布を知る上で極めて有用なプローブを得ることが出来た。
