CD3合成酵素遺伝子により誘導される細胞増殖シグナルの機構解析

研究概要	ヒトのメラノーマやT細胞白血病に特異的に発現するGD3の細胞増殖における意義につき、GD3合成酵素遺伝子を種々の細胞に導入して、その表現型の変化を検討することにより解析した。まずラット褐色細胞腫PC12に、GD3合成酵素遺伝子を導入したトランスフェクタントでは、1.著しい増殖速度の亢進、2.神経成長因子(NGF)による分化誘導に対する不応、3.無血清処置によるアポトーシスへの抵抗性の獲得などが主な変化として観察された。著しい増殖能をもたらしたメカニズムの検討のため、NGF受容体TrkAのリン酸化、その下流のMAPキナーゼの活性化状態につき検討したところ、GD3合成酵素遺伝子導入細胞では、NGF刺激の有無に関わらず、TrkAのリン酸とMAPキナーゼの活性化を認めた。種々のキナーゼインヒビターを用いた増殖抑制実験の結果から、このトランスフェクタントで見られる増殖の亢進がTrkA/Ras/MAPキナーゼ系を介していることが示唆された。また、TrkAの二量体形成もNGF刺激に無関係に見られた。よって遺伝子導入によりTrkAの構造的変異が惹起され、恒常的な活性化状態を招いたと思われる。また、無血清培地によるアポトーシス誘導に対して、このトランスフェクタントは抵抗性を示したが、PI3キナーゼインヒビターのワルトマニン存在下ではアポトーシス抵抗性が消失した。よって、このトランスフェクタントでは糖鎖変化によってPI3キナーゼ活性が上昇していることが示唆された。これらの結果は、メラノーマやT細胞白血病の腫瘍増殖の機構の一部を反映するものと思われ、現在メラノサイトに対する遺伝子導入、糖脂質改変を行って、その細胞変化の検討を行っている。
