マウス乳癌細胞増殖・転移におけるテネイシンの生物作用の研究

研究概要

1)抗テネイシン(TN)抗体によるヒト癌細胞の腫瘍oェル成長における
ヒト癌細胞を用いて抗TNl功区抗TN抗体の腫瘍成長抑制効果を検討した。TNの異なるドメインを認識する単抗体(EGF様配列を認識するRCBl、DEARl、AD配列を認識するDEAR2など)を皮下移植した癌細胞に投与した。細胞は培養下でTN非産生細胞株のA431を用いた。その結果、各抗体ともに腫瘍成長を抑制し、特に、RCBlの抑制効果が高かった。従って、TNのEGF様配列がヒト癌細胞増殖に対しても重要であることが確認された(第57回日本癌学会、横浜、1998年)。
2)テネイシン誘導因子(TIF)誘導活性および細胞増殖への影響マウス胎仔線維芽細胞の培養上清中にはTIFが存在することを見出した。TIFのTN誘導活性を他のサイトカインと比較したところ、TGF-βやEGFに比べてさらに強い誘導活性を示した。各サイトカインよる刺激後、チロシンリン酸化の有無を調べたところ、TIFはEGFやTGF-αと異なり、チロシンをリン酸化しないことが示された。次に、マウス乳癌細胞(GLMT1)にTIFを添加したところ、経時的にTN量は増加し、同時に、細胞増殖の促進も見られた。しかし、TN欠損マウス由来乳癌細胞にTIFを添加しても、細胞増殖は変わらなかった。従って、TIFの細胞増殖活性は、誘導されたTNを介していると思われる(第8回乳癌基礎研究会、鳥羽、第22回国際乳癌学会、アテネ、1998年)。
3)TINに対する単抗体の作製
TIF活性画分を抗原としてラットを免疫し、ラット/マウスのハイブリドーマを作製、1gGの産生とTIFの中和阻害活性を指標としたバイオアッセイにより抗TIF抗体産生株の候補を選択しクローニングを行った。現在までに11クローンが得られ、細胞内局在と生体組織内における局在を免疫組織学的に検討中である。
