ポリアミン代謝拮抗に基づく抗がん剤の分子設計

研究概要	1. eIF5Aは細胞内でポリアミンが共有結合して存在する唯一の蛋白質であり、生存に必須の因子である。このeIF5Aの生合成をデオキシスパーガリンが阻害し、細胞増殖阻害が起こることを明らかにした。デオキシスパーガリンはスペルミジンがハイプシンとしてeIF5Aに結合するための律速酵素であるデオキシハイプシン合成酵素の阻害剤であること、またデオキシスパーガリン中のグリオキシスペルミジン部分が阻害に関与していることが明らかとなった。細胞増殖阻害に関しては、オルニチン脱炭酸酵素(ODC)の阻害剤であるα-ジフルオロメチルオルニチンとの併用を試みたが、相乗効果は認められなかった。 2. アンチザイムはODCの分解を速めるだけでなく、ポリアミン輸送も負に調節した。ポリアミン輸送の負の調節は取り込みの抑制と排出の促進の両者に依存していた。このことはアンチザイムが直接ポリアミン輸送蛋白質に結合することにより起こると推定された。アンチサイム遺伝子をがん細胞にトランスフェクトして抗がん効果を調べたところ、強い抗がん効果が認められた。 3. 酵母のポリアミン輸送欠損株を用いて真核細胞のポリアミン輸送蛋白質遺伝子のクローニングを試み、TPO1(transporter for polyamine 1)遺伝子のクローニングに成功した。この遺伝子は12膜貫通領域を有する液胞膜上の蛋白質をコードし、この蛋白質は基質としてポリアミン(プトレスシン、スペルミジン、スペルミン)のみを認識した。
