ホルモン依存性生殖器癌発症の基礎的研究

研究概要	乳癌、子宮体癌や前立腺癌などの性ステロイドホルモン依存性生殖癌は、本邦において増加の一途をたどっている。本研究では、以下の2点に焦点を当て、性ステロイドホルモン作用発現における鍵分子である核内ステロイドレセプター及びその転写共役因子群に絞り、その基本的な性質を明らかにするとともに、新たな転写共役因子の検索および性状解析を進め、発癌の過程で生じる異常について考察した。 1.既知核内レセプター転写共役因子群のERα、ERβへの効果:転写制御因子である核内レセプターは、2つの転写促進領域をレセプタータンパクN末端とC末端に有する。しかしながら、ホルモン結合によるレセプターの構造変化とそれに関わるこれら2つの領域の機能的な相互作用や、これに関与する転写共役因子との相互作用は全く不明である。そこで、既知転写共役因子の相互作用の解析を行った結果、p300がERα、ERβのN末端、C末端共に作用し、機能を橋渡しすることがわかった。 2.ホルモン活性を規定する核内レセプター転写共役因子の同定:上述した転写共役因子として既に同定されたものには、転写を促進するco-activatorと転写を抑制するco-repressorの存在が知られている。そこで性ステロイドレセプター種[ER、アンドロゲンレセプター(AR)]固有の転写共役因子の検索・同定を、酵母two-hybrid法を用いたcDNAスクリーニング、及び生化学的手法を用いて行い、候補因子をいくつか同定した。そのうちDEAD-boxを有するp68RNAヘリケースとRNA結合タンパク質p72がERαAF-1の転写共役因子として働くことを見出した。さらに、p72がco-activatorとして知られるRNA分子であるSRAと協調的にERαAF-1活性を亢進することを明らかにした。
