T細胞認識腫瘍抗原の解析と腫瘍ワクチン

研究概要	BALB/cマウス白血病RL♂1には、特異的CTLが認識するAkt分子由来の拒絶抗原ペプチドpRL1aが見出されている。しかし、BALB/cマウスではRL♂1の拒絶は起こらない。われわれはCD4T細胞がAktを認識しCD8T細胞のpRL1aに対する有効な免疫反応を阻害していることを明らかにした。また、CD4^+CD25^+T細胞は、腫瘍抗原非特異的に拒絶反応を阻害していることも明らかにした。一方、pRL1aの多価ペプチドによる免疫効果をイムノモニタリング法としてエリスポットアッセイを用いて明らかにした。さらに、BALB/cメチルコラントレン誘発線維肉腫MethAに対する宿主液性免疫反応をSEREX法により検討した。MethAマウスにIFN-_γ cDNAを導入したMethA(K_γ)株からcDNAライブラリーを作成し、これを親株MethA担癌BALB/cマウスから得た血清でスクリーニングし、35個のクローンを得、塩基配列の検討から10個の抗原を同定した。このうちOY-MS-4は、X染色体上にある胎盤・胎児発現遺伝子pemと同一であった。OY-MS-4は、正常組織では、精巣・胎盤のみに発現し、腫瘍では、検索した15種類の株のうち10種類に発現していた。OY-MS-4は新しいC/T(cancer/tistis)抗原である。OY-MS-4以外の他の9つの抗原は正常組織では種々の発現パターンを示し、腫瘍では、調べた15種類すべてに発現していた。10個の抗原はMethA担癌マウスにのみ抗体が検出され、他の腫瘍については抗原を発現しているにも拘らず、それらの担癌マウスに抗体は検出できなかった。塩基配列には突然変異は認められず、共通抗原でありながら固有抗原性を示すことが明らかになった。
