発達期小脳における登上線維シナプス除去の臨界期と神経活動依存症に関する研究

研究概要	成熟動物では小脳プルキンエ細胞はただ1本の登上線維によって支配されるが、発達初期には3〜5本の登上線維の支配を受ける。発達につれて過剰な登上線維が淘汰され、生後約20日で成熟型の1対1の結合が完成する。先行研究から、NMDA受容体の関与が示唆されていたが、この過程が神経活動に依存するかについての直接的な証明がなく、また、小脳生後発達のどの時期に必要であるか(臨界期)については明らかでなかったので、本研究においてこの2点を追求した。 (1) 神経活動依存性の検討:テトロドトキシン(TTX)をethylene-vinylacetate copolymer(ELVAX)に溶媒によって混合し、その微小ブロックを生後10日の発達期マウス小脳の表面に移植して、これからTTXを持続的に小脳に与えた。マウスを成長させ、生後24日から36日の間に、登上線維の支配様式を電気生理学的に解析した。ELVAX近傍の小脳6-8葉で記録すると、vehicle投与群に対してTTX投与群では、登上線維による多重支配を受けるプルキンエ細胞の割合が有意に高かった。これに対し、ELVAXから離れた小脳1/2および10葉では、両群間で有意差はなかった。したがって、TTXの効果は小脳内に限局しており、小脳内の神経活動が登上線維シナプス排除に必要であると考えられた。 (2) 臨界期の検討:NMDA受容体の特異的阻害剤のMK-801(0.25μg/g体重)を1日1回生後7日から14日まで8日間腹腔内投与し、その後投与をやめて生後25日まで成長させた群(P7-P14投与群)、生後15日から21日まで7日間腹腔内投与した群(P15-P21投与群)および生理食塩水投与対照群で比較したところ、P15-P21投与群でのみ登上線維シナプス排除に異常を認めた。この結果は、登上線維シナプス排除の臨界期は生後15日以降であることを示す。
