新奇な神経特異プロテオグリカンNGCのバレル形成における機能の解析

研究概要	NGC(ニューログリカンC)は、私達が発達期のラット脳において発見した新奇な膜貫通型コンドロイチン硫酸プロテオグリカンである。本研究では、脳の神経回路形成におけるNGCの役割を解明することに、重点を置いている。本年度は、次の諸点を明らかにした。 1. マウスのNGCゲノムDNAの構造 NGCの発現調節機構の解析と、ノックアウトマウスの作製を目的として、マウスNGC遺伝子の構造を調べた。その結果、約17kbの範囲に、6個のエクソンが存在することが明らかとなった。また、NGC遺伝子(CSPG5として国際登録)は、マウスでは9番染色体(9F1)に存在することがわかった。 2. 形成途上のバレルにおけるNGCおよびその関連分子の局在 NGCは、細胞外領域に、上皮増殖因子(EGF)様のドメインを持つことから、EGFファミリー分子と考えられる。脳に存在する代表的なEGFファミリー分子としては、ニューレグリンが良く知られている。そこで、発達期のラット大脳体性感覚野におけるNGCとニューレグリンの分布を、免疫組織化学的に調べた。NGCはバレルの隔壁に存在するのに対して、ニューレグリンはバレルの内部に検出された。バレル内部には、EGF受容体であるErbB2とErbB4も検出できることから、ニューレグリンはこれら受容体と作用し合って、視床-皮質線維と皮質神経細胞とのシナプス形成を促進している可能性がある。一方、形成途上のバレルの隔壁に存在するNGCは、視床-皮質線維とNGC発現細胞とのシナプス形成を阻害することにより、バレルという脳の機能単位の形成に寄与していると思われる。
