雌雄異株植物を用いた性染色体と生殖器官特異的遺伝子群に関する分子細胞学的解析

研究概要	本研究の目的は、性染色体をもちXY型の性決定を行う雌雄異株植物メランドリウム(ヒロハノマンテマ、Silenelatifolia)を用いて生殖器官形成に関わる遺伝子群を単離・同定すると同時に、高等植物の性染色体にコードされる遺伝子群が生殖器官形成に果たす役割を明らかにすることである.特に、雄にしか存在しないY染色体は性決定において重要な役割を担っていると考えられるので、本研究では雄性生殖器官特異的に発現される遺伝子群およびY染色体特異的遺伝子群に解析を集中した.今年度の成果は以下のとおりである. (1) レーザーマイクロダイセクションを用いて単離されたY染色体由来の高度反復配列のひとつ、RMY1(repetitive sequence in microdissected Y chromosome)は、チェコのグループがマイクロダイセクションによりメランドリウムX染色体から単離したサプテロメア様配列X43.1に高いホモロジーを示した.FISH解析の結果、RMY1は常染色体を含む多数の染色体末端部分に局在を示したが、性染色体間では局在様式が異なっていた(X染色体は両端、Y染色体は一方の末端).この配列は性染色体の異型化を考察する上で有用な指標になると考えられる. (2) 10merのプライマーを用いて雌雄ゲノムDNAを鋳型とするRAPD解析を行い、雄DNAから特異的に増幅されてくるDNA断片を得た,ゲノミックサザンハイブリダイゼーションにより雄特異性を検討し、最も雄特異性が高いと思われたクローンについてSTS化を行い、雄特異的に単一のバンドが増幅されることを示した.inverse PCRにより、その周辺領域の取得を試みたところ、逆転写酵素様のタンパク質をコードし得るORFを含む約9.6kbpのクローンを得た.この配列はY染色体DNAのマーカーとして有用であるとともに、Y染色体特異的遺伝子の有力な候補である.
