ヒトT細胞白血病ウイルスによる自己免疫発症機構の解析

研究概要	ヒトの関節リウマチとよく似た関節炎を発症するHTLV-I Tgマウスの関節で増殖しているT細胞の抗原特異性を調べたところ、II型コラーゲンに反応するものが見つかった。HTLV-I Tgマウスは関節炎を自然発症するだけでなく、コラーゲン誘導関節炎(CIA)に感受性になっていることからも、II型コラーゲンに対するトレランスが破綻している可能性が示唆された。次に関節炎発症におけるサイトカインの役割を明らかにするためにHTLV-I Tgマウス及びCIAに高感受性のDBN/1マウスと、種々のサイトカインKOマウスとの掛け合わせを行い、発症率を比較した。どちらの系においてもIL-1とIL-6の炎症性サイトカインが発症に重要な役割を果たしており、TNF-αの欠損は発症率に影響を与えなかった。しかしHTLV-I TgではIL-1KOやIL-6KOマウスでも最終的にはある程度発症したため、これらのサイトカインの欠損は他のサイトカインで相補されることが示唆された。一方、Th1とTh2のバランスに重要なIFN-γとIL-4の欠損は発症率に影響を与えないことが明らかとなった。CIAの発症に重要な役割を果たしているII型コラーゲンに対する抗体産生能をこれらのサイトカインKOマウスについて調べたところ、IL-6KOマウスにおいて有意に抑制されており、このことが発症を抑制している可能性が考えられた。しかし、HTLV-I TgのモデルではIL-1KOもIL-6KOマウスも発症したものでは自己抗体がコントロールと同じレベル産生されており、これらのサイトカインは抗体産生とは異なる作用で発症に関与している可能性が示唆された。
