炎症性腸疾患(IBD)発症と修復におけるIELの機能解明

研究概要	潰瘍性大腸炎やクローン病などのヒト炎症性腸疾患(inflammatory bowel disease:IBD)の発症基盤に腸管免疫の異変、主として腸管T細胞の異常機能のあることが明らかにされている。昨今では各種サイトカインやT細胞抗原受容体(TCR)遺伝子欠損マウスが高頻度にヒトIBDに酷似した腸疾患を発症することが報告されている。腸管T細胞の大多数は一層の粘膜上皮細胞間に分布する腸管上皮細胞間T細胞(intestinal intraepithelial T lymphocyte:IEL)であるが、これらはいわゆる胸腺で発達分化して末梢リンパ組織に分布するT細胞とは異なっており、腸管粘膜局所で発達分化することが明らかにされている。 TCR-α鎖欠損(α^<-/->)マウスはTC8-P鎖欠損(β^<-/->)マウスとは対照的にIBDを高頻度に発症するが、本研究によってα-l-マウスではIBD発症以前からγδ-IELのキラーT細胞活性やIFNγ産生能が異常に亢進することが明らかにされた。さらに腸内常在細菌の存在しない無菌のα^<-/->マウスにはIBD発症が見られないことから、腸内細菌がγδ-IELを活性化することがIBD発症の基盤と考えられる。すでにα^<-/->マウスにおけるIBD発症の責任細胞はTCR-ββホモダイマーを発現しTh_2タイプのサイトカインを産生するββT細胞であることが明らかにされていることから、γδT細胞の機能亢進によってββ細胞の活性化が引き起こされる可能がある。今後、α^<-/->マウスのγδ-IEL機能亢進に関与する腸内細菌の同定を目ざしたい。
