ヒト・ゲノムの機能的ドメイン構成の解析ならびにFISHの技術開発

研究概要

ヒト・ゲノム上に存在する何万もの遺伝子は、ゲノム上で均一に配置されているのではなく、それぞれの機能に応じてクラスターを形成するなど、機能的ドメインを形成し制御を受けていることは明らかである。本研究では、ゲノム全体をDNA複製やダイナミックなゲノムの伸縮構造に基づく機能的、構造的ドメインによる構成体として捉え、これらのドメイン構成を明らかにするとともに、細胞核内のゲノムの配置や挙動を解析することにより、ゲノムの構成を総合的に理解し、ゲノムの機能研究として、生命現象の基盤である遺伝子の精妙な機能発現の理解に資することを目標とした。まず、DNA複製前の核内のゲノムのパッケージング状態を、halo核標本に対してFISHを行うことにより解析し、インプリント遺伝子SNRPNでは発現している父親由来アレルがコンパクトにパッケージングされていることを示した。また、イントロンプローブやゲノムクローンをプローブとして核内RNAの検出を試み、インプリント遺伝子の単一アレルでの発現を確認した。この方法により、非同調的複製パターンを示すDiGeorge領域のクローンを検討したところ、biallelicに発現していることを確認した。さらに、複製鎖の蛍光検出と特定ゲノムに対するファイバーFISH法を組み合わせることで、複製部位を個々の細胞で確認する方法を開発した。本年度は特に、複製鎖やFISHが安定なシグナルを与える方法の改良を行い、両者の同時検出にも成功した。また、セントロメアへテロクロマチン領域の低アセチル化状態の維持機構として、別の場所で脱アセチル化されたヒストンが、セントロメア領域に輸送されるという機構が存在する可能性を提示した。さらに、各種遺伝子、トランスジーンのマッピング、ゲノムの整列化を行った。
