機能探索分子の活用によるニコチン性受容体を介して発現される神経保護作用機構の解明

研究概要

1) 神経保護作用を有するニコチン性受容体サブユニットの決定:初代培養大脳皮質神経細胞において、ニコチン性受容体刺激-による神経保護作用の少なくとも一部はα4β2サブユニットを介して発現することを明らかにした.
2) ニコチン及びその関連物質のβシート構造形成に対する効果の検討:蛍光色素であるthioflavin Tを用いてβシート構造形成について再検討した。いずれのAβ fragmenβシート構造を形成した。コンゴレッドは著明にその構造形成を減少させたが、ニコチンは影響を与えなかった。また、βシート構造の形成過程に対しても、ニコチンは影響を与えなかった。ニコチンの神経保護効果は、βシート構造形成を減少させることで発揮されるという説は我々の系では否定された。
3) ニコチン受容体刺激とPI3キナーゼ系との関連:Aβ神経毒性は、グルタミン酸毒性に対するニコチンの保護作用は、PI3キナーゼの拮抗薬であるLY294002により抑制された。PI3キナーゼによりリン酸化されるAktのリン酸比を認めた。PI3・キ+ーゼは細胞生存に関わると考えられ、ニコチン受容体刺激による神経保護効果がこの系を介する可能性が示唆された。
4) ドパミン受容体刺激とグルタミン酸神経毒性:ドパミンD2受容体の刺激薬であるブロモクリプチン、キピロールはグルタミン酸毒性を抑制した。さらにカルシウムイオノフォア、NO生成試薬である.NOC18やperoxynitrite生成試薬であるSIN-1により誘発される神経細胞死抑制も。抑された。と.の効果はD2アンタゴニストで抑制された。さらにD1 アゴニストでは神経保護効果は認めなかった。D2アゴニスの神経保護効果はLY294002により抑制された。またAktのリン酸化を終めた。D2受容体刺激による神経保護効果がPI3キナーゼ系を介する可能性が示唆された。
