PHドメイン結合物質によるPKB蛋白質リン酸化酵素の活性制御に関する研究

研究概要	-PKBはアミノ末端側にPHドメインを持ち、カルボキシル末端側に触媒ドメインを持つセリン/トレオニン蛋白質リン酸化酵素であり、細胞増殖因子刺激によりホスファチジルイノシトール3-キナーゼの下流標的として活性化されることが提唱されている。私どもは熱ショック、高浸透圧等のストレスによりPKBが活性化を受けることを明らかにしている。熱ショックによるPKBの活性上昇はホスファチジルイノシトール3-キナーゼ阻害剤により抑制を受けず、また、PHドメインの欠失変異分子では活性化は生じないことから、ストレス刺激によるPKB活性化にはPHドメインにホスファチジルイノシトール3-キナーゼ反応産物以外の物質が作用していると推定される。そこで、PKBのPHドメインに特異的に結合する蛋白質の検索を行なった。その結果、培養細胞抽出液中にPKBのPHドメイン-GST(glutathione S-transferase)融合蛋白質に特異的に結合する分子量約200kDの蛋白質が見い出され、各種の抗体を用いた、イムノブロット法によりこの蛋白質はミオシンII重鎖と同定された。次に細胞内でのPKBとミオシンIIとの結合を検討した結果、PKBのPHドメインのみを発現した場合は恒常的にミオシンIIとの結合が観察されたが、全長PKBとミオシンIIとの結合は検出されなかった。従って、PKBのPHドメインは細胞内においてもミオシンIIと結合しうるものの、PKBとミオシンIIとの結合には何らかの細胞刺激が必要であると推測される。一方、熱ショックにより細胞内でPKB活性化に伴いPKB同士がそのPHドメインを介して会合しオリゴマーを形成することが明らかとなった。以上の結果から、PKBはそのPHドメインを介する特異的結合蛋白質との結合およびPKB同士の会合を介してその細胞内局在や蛋白質リン酸化酵素活性が制御を受けていると考えられる。
